Functional characterization of Toll-like receptor 4 (TLR4) in salt-sensitive hypertension. Toll-like receptor 4 (TLR4) has been implicated in the pathogenesis of hypertension. Hypertension may be induced by various environmental factors, including high salt intake. However, the role of TLR4 in salt-sensitive hypertension remains unclear. In the present study, a rat model of salt-sensitive hypertension was used to determine the functional role of TLR4 in this model. Male Dahl salt-sensitive (DS) rats and Dahl salt-resistant (DR) rats were maintained on 0.3% or 8% NaCl diets. Blood pressure was measured with a tail-cuff method in conscious rats. Protein expression levels of TLR4 were determined by Western blot analysis, and TLR4 functional activity was assessed by transient expression in HEK293T cells. Blood pressure was significantly higher in DS rats than in DR rats on either diet. The expression level of TLR4 was significantly higher in DS rats than in DR rats, irrespective of diet, and the TLR4 expression level was higher in DS rats than in DR rats on the high-salt diet. A time-dependent increase in TLR4 mRNA was detected in cultured aortic smooth muscle cells from DS rats on the high-salt diet, and this increase was inhibited by the inhibition of NF-κB activation. A TLR4 agonist, LPS, induced the elevation of blood pressure in DR rats.
